Dr. David Clarke's research group focuses on the application of modern techniques in mass spectrometry (MS) for the functional and structural characterisation of biological systems. The research utilises a range of MS-based techniques including - top-down and middle down fragmentation, ion-mobility mass spectrometry, native mass spectrometry, and hydrogen/deuterium exchange. In addition, MS-based studies are supported by complementary biochemical analysis and structural biology approaches.

The Implementation and Impact of Isotope Depletion for Improved Protein Mass Spectrometry.
One of the fundamental challenges of high-resolution mass spectrometry (MS) analysis of protein occurs as the molecular weight increases. Which is due to the increased chemical complexity of the protein analyte. Larger ... -
Mass Spectrometry Analysis of Encapsulated Ferritins from Haliangium ochraceum – Native IM-MS and HDX-MS
Encapsulated ferritins (EncFtn) are a recently characterised member of the ferritin superfamily. EncFtn are stored within encapsulin nanocompartments and together, they form an iron storage system. Here we elucidate the ... -
Untargeted Metabolite Mapping in 3D Cell Culture Models using High Spectral Resolution FT-ICR Mass Spectrometry Imaging - Datasets
Mass spectrometry imaging (MSI) is an effective tool to investigate multicellular tumor spheroids (MTS) in a spatially-resolved manner. It has been used to study the distribution of lipids, post-translational modifications, ... -
Characterisation of enzymatic processing of commercial (Kraft) lignin by high resolution FT-ICR mass spectrometry – Dataset.
High Resolution FT-ICR mass spectrometry datasets analysing the enzymatic processing of Kraft lignin. -
Isotope Depletion Mass Spectrometry (ID MS) for Enhanced Top-Down Protein Fragmentation - Datasets
Top-down mass spectrometry has become an important technique for the identification of proteins and characterisation of chemical and posttranslational modifications. However, as the molecular mass of proteins increases ... -
Native MS Analysis of Encapsulated Ferritins from Rhodospirillum rubrum, Haliangium ochraceum and Pyrococcus furiosus
Intact mass spectrometry (LC-MS) and native mass spectrometry analysis of 3 homologs of the encapsulated ferritin enzyme. This data is used in the study: 'Conservation of the structural and functional architecture of ... -
Optimization of sprayer for matrix application in MALDI mass spectrometry imaging
Successful matrix-assisted laser desorption ionization (MALDI) mass spectrometry imaging (MSI) relies on the selection of the most appropriate matrix and optimization of the matrix application parameters. In order to achieve ... -
ESI-(-)-FT-ICR MS of 85 Scotch Whisky Samples
Data were acquired as part of research into advanced spectroscopic methods for the analysis of Scotch Whisky. 85 Scotch Whisky samples, representing the broad range of Scotch Whisky produced and sold, were analysed by ... -
Top-Down Protein Electron Capture Dissociation Mass Spectrometry Datasets
Top-down electron capture dissociation (ECD) fragmentation datasets for several widely available commerical protein samples. All data were collected on a 12T SolariX FT-ICR mass spectrometer (Bruker Daltonics); and data ... -
Native MS Analysis of Encapsulated Ferritin from Rhodospirillum rubrum
This dataset contains native MS analysis of the Encapsulated Ferritin (EncFtn, Rru_A0973) from Rhodospirillum rubrum. Included are ion-mobility mass spectra of apo- and Fe-bound EncFtn, mass spectral data showing the gas ...
